I am a listing broker who agreed to pay the cooperating broker $5000. The buyer’s agent has presented the seller with an offer asking for seller concessions of $7500. Is the commission amount included in the $7500?

No. If your seller accepts this offer, they will be required to contribute $7500 toward closing costs IN ADDITION TO the compensation that you have already agreed to pay the buyer’s agent. Be very careful when reviewing offers!
